Overview
Epitalon is among the longest-observed peptides in longevity science. It is a short, fully synthetic molecule built from just four residues (alanine, glutamic acid, aspartic acid, glycine, abbreviated AEDG) whose composition mirrors the amino acid balance of Epithalamin, an extract of the pineal gland. It was designed by the team of Vladimir Khavinson, based at the Institute of Bioregulation and Gerontology in St. Petersburg, and first synthesized in the year 2000.
What the literature has recorded so far:
- Telomerase in somatic cells: Human fibroblasts that normally stop dividing around passage 34 kept going past passage 44 under Epitalon and held on to their telomere length. In cells that do not make the enzyme at baseline, the catalytic subunit was switched on, enzyme activity became measurable and the telomeres grew.
- Pineal gland and rhythm: Animal models show a melatonin profile back in the physiological range, a normalized day-night cycle and signs of protection for nerve cells.
- Further signals: Changes in interleukin-2 mRNA, influence on the enzymes AChE and BuChE, and geroprotective findings across several animal species.
A 2025 paper refined the picture: in normal epithelial and connective tissue cells the telomere gain ran dose-dependently through hTERT and telomerase, whereas breast cancer cell lines lengthened their telomeres via the ALT route instead. That puts Epitalon among the very few substances with a documented telomerase switch-on in ordinary human body cells. Study protocols mostly work with short blocks of roughly ten days rather than continuous dosing; the models used subcutaneous or intramuscular routes.
Risk notes
Tolerability is described as good in the literature; across several decades of the Khavinson program no notable toxic effects were recorded. Still, there are points you should know:
- Local: The injection site may redden, feel tight or become briefly irritated. This is by far the most frequent report.
- Circulation: Occasionally a short spell of tiredness or a mild sensation of warmth is mentioned shortly after dosing.
- One-sided data base: The lion's share of the studies comes from a single institute; independent replications from other countries are scarce. The effect on telomerase in particular leaves mechanistic questions that nobody has conclusively answered yet.
- No long-term human data: What is known about safety comes from animal work and short observation windows; multi-year human studies do not exist.
