Overview

Mazdutide takes a native hormone as its template: oxyntomodulin, which in humans engages both the GLP-1 and the glucagon receptor. The synthetic analog, also listed in the literature as IBI362 or LY3305677, carries over exactly this double profile.

Origin and development status:

  • Developed jointly by Innovent Biologics and Eli Lilly; development in China is led by Innovent.
  • Phase 2: up to 15.4 percent less body weight after 24 weeks.
  • Phase 3: Chinese market data are on the table, and the substance was approved for use there in 2025.

What the two receptors contribute: through GLP-1, research models see appetite regulation addressed; through glucagon, higher energy expenditure. Together that yields the typical signature of the GLP-1/glucagon class.

Pharmacokinetics from the trials:

  • Half-life in the range of about seven days.
  • Peak plasma level typically around 72 hours after administration.
  • Hence one subcutaneous administration per week in the protocols.

Within its class, mazdutide is one of the few molecules modeled this directly on the natural hormone. For comparative research that uses oxyntomodulin itself as the reference, that is an advantage in its own right. Below you find the phase 1b dose study, the phase 2 obesity trial and the phase 2 work in type 2 diabetes.

Risk notes

According to the studies, mazdutide's tolerability matches that of a GLP-1/glucagon dual agonist: acceptable overall, shaped by the gut and, in the phase 2 data, dependent on dose.

  • Stomach and bowel: nausea, vomiting, diarrhea and constipation top the list, mostly at the start and with rapid dose increases; slower escalation lowered the frequency.
  • Fluids: the dampened appetite can cut how much is eaten and drunk; the literature describes a higher risk of dehydration.
  • Heart rate: mild increases are documented for the glucagon component.
  • Muscle and injection site: part of the weight loss falls on muscle mass, and local irritation at the injection site is described.
  • Evidence base: most of the data come from China, and there are no robust figures yet on very long use.
