Overview

If you look PT-141 up in the literature you will mostly find it under the name bremelanotide. It is a descendant of melanotan 2: a ring-closed peptide of seven amino acids that docks onto melanocortin receptors. Where its parent compound drives pigment formation, PT-141 is dominated by a different pathway, the activation of MC3R and MC4R, with the MC4R pathway inside the brain as the actual target.

What the research shows so far:

  • In studies with men and women, measures of sexual arousal and desire changed after administration.
  • In 2019 the compound was approved in the US for premenopausal women with hypoactive sexual desire disorder, so more controlled data exist than for most other peptides.
  • Further work looks at melanocortin-dependent pathways of appetite regulation and the stress response.

On the mechanism: the effect does not arise in the blood vessels but centrally in the hypothalamus, where melanocortin receptors help steer the sexual response system. This sets PT-141 apart in principle from compounds that act on vascular tone.

Pharmacokinetics from the studies: after subcutaneous administration the peptide is well absorbed, plasma levels peak after roughly one hour, and the half-life is two to three hours. Elimination runs mainly through the kidneys, and binding to plasma proteins is low.

We list PT-141 strictly as a research compound. What you read here places the evidence in context and is not a guide to use.

Risk notes

The approval trials documented a clear pattern of side effects, most of it traceable to central melanocortin activation:

  • Nausea comes first and increases with the dose.
  • Facial warmth, flushing and headache are reported regularly.
  • Blood pressure can rise temporarily while heart rate drops slightly; both resolved in the trials.
  • Because pigment cells also carry melanocortin receptors, skin can darken with repeated administration, as can moles and freckles.
  • Redness and tenderness at the injection site are common.
