Overview

This blend brings together two peptides that both come from the Institute of Molecular Genetics in Moscow and are approved in Russia: Selank and Semax, in equal parts in one solution. Both are short chains of seven amino acids; Selank reads Thr-Lys-Pro-Arg-Pro-Gly-Pro, Semax Met-Glu-His-Phe-Pro-Gly-Pro. The appeal of the pairing is that, in the literature, the two activity profiles complement each other rather than overlap.

Selank, the calming half:

  • modulates GABAergic and serotonergic signalling
  • shows anxiolytic effects in study models without sedation
  • changes gene expression in the rat hippocampus, after a single dose as well as after repeated dosing
  • raises BDNF in the hippocampus after nasal dosing

Semax, the activating half:

  • in the 2006 rat study, increases BDNF and the phosphorylation of the receptor TrkB in the hippocampus
  • shifts cholinergic and dopaminergic signalling, which is linked to cognitive gains
  • in a stroke model, modulates genes of the immune and vascular systems, shown in a genome-wide analysis

In practical terms, the ready-made mixture saves two separate solutions; in studies, both peptides are given nasally. What does not exist is data on the combination itself: every statement about effect and tolerability comes from work on the individual substances, nearly all of it from Russian groups. Whether the effects add up, weaken or mask each other has not been studied systematically so far.

For interpretation this means: the Selank literature carries the statements on anxiety relief, the Semax literature those on cognition, and the two have to be read separately.

Risk notes

After decades of Russian research, both components are rated low in side effects. What you should keep in mind with the mixture:

  • Nasal mucosa: burning, dryness or irritation is the most common observation.
  • Two directions in one vial: Selank calms, Semax activates; depending on the time of day, both tiredness and mild restlessness are therefore described.
  • Activation: the Semax share engages stimulating and cognitive pathways; pressure in the head or a shifted sleep pattern are named in the Semax literature, mostly after late dosing.
  • No clean attribution: which part triggers which effect cannot be told apart in a mixture.
  • Origin of the data: most studies on both peptides come from Russian groups, and independent confirmation is rare.
  • Data situation: long-term studies outside Russia are largely missing, and investigations of the combination as such do not exist.
